Sucrose gradient purified Human Herpesvirus 6 Type B (HHV-6B), strain Z-29 viral DNA for PCR. Extract preparation/DNA quantitation is SDS, proteinase K digestion followed by phenol/chloroform extraction and ethanol precipitation. DNA concentration is determined by fluorometry using Pico Green. PCR analysis was performed on purified viral DNA using the Applied Biosystems Core Reagents PCR Kit and primers specific for the HHV-6 DR-2 region, which differentiate between HHV-6 types A & B by fragment size.
