Purified virus made from clarified, virus infected tissue culture harvest fluids that are concentrated and cleaned of debris by a combination of ultracentrifugation, ultrafiltration and selection of viral band from a gradient. This preparation contains live virus and can be used to transform primary human lymphocytes.

Transformation Assay: The transformation dose 50 (TD50) of purified EBV B95-8 was performed using gradient separated adult peripheral blood mononuclear cells (PBMCs). The PBMCs were inoculated with 0.2 mL/well log10 dilutions of EBVB95-8 virus in 4 replicates per dilution and incubated at 37°C for three weeks in RPMI 1640 + 10% FBS. Cultures were refed and examined microscopically for aggregate/clusters of transformed proliferative B-lymphoblast cells and the end-point for transformation read at the end of three weeks by microscopic examination for the presence of transformed clusters of cells. The transformation endpoint was confirmed by intranuclear fluorescent-stained granules with mouse monoclonal antibody to EBNA-1 (ABI Cat. No. 13-156-100) and the TD50/mL calculated using the Karber method.
