Validation of Inactivation: Beta-Propiolactone (β-PL) is a DNA alkylating agent that has been shown to be effective in the inactivation of viruses. B-PL chemically modifies viral DNA without affecting the immunogenicity or protein structure. After treatment with β-PL, infectious HCMV, strain AD169 was completely inactivated as validated by cell culture based assay (active HCMV, strain AD169 had a TCID50 titer of 108.0 TCID50/mL), but retained viral morphology (by TEM).
